so /backup have no space to take backup. All free space allocated to /home.

so can i use a backup folder like this /home/backup

Yes, that is fine. You've to create the folder /home/backup first, and then enter this in the cPanel Backup Configuration settings to use it going forward during backup generation. I'd also suggest keeping backups in remote storage too.
